Study on diamine Oxidase Activity in Human Amniotic and Chorionic Tissues


Abstract
The activities of diamine oxidase in cytoplasm, mitochondria and nuclear fractions isolated from human amniotic and chorionic tissues were measured together with some biochemical characteristics and km values -to substrates.
The results were summarized as follows:
1. Diamine oxidase activities in amniotic and chorionic tissues were 46.95 and 47.38 units
/g of wet tissue, respectively.
2. The specific activity of diamine oxidase in amniotic tissue was twice as hi-h as that in chorionic
Y~ tissue. (8. 6lunits/mg protein: 3. 46 units/mg protein)
j` 3. Diamine oxidase was distributed 79. 2io in cytoplasm, 10. 8% in mitochondria and 10.0% in nuclear fractions of amniotic tissue.

?f Y fractions of chorionic tissue.
5. Activity of diamine oxidase in amniotic tissue was different according to substrates: 36.050.93 units
Y /g of wet tissue in putrescine, 45. 563.04 units/g of wet tissue in cadaverine, 63.584-3. 34 units/g of
¢¥ wet ¢¥tissue in histamine, 40. 81+-1.48 units/g of wet tissue in spermidine and 41.290. 02 units/g of
wet tissue in spermine. The activity was the highest in histamine and was the lowest in putrescine.
6, Activity of diamine oxidase in chorionic tissue was different according to substrates: 54.350.95 units/g of wet tissue in spermine; 45. 550. 62 units/g of wet tissue in cadaverine, 37. 41--0. 44 units/g of wet tissue in putrescine, 35. 640.62 units /g of wet tissue in spermidine and 31. 35+0, 71 units/ g of wet tissue in histamine. The activity was the lowest in histamine.
7. Km value of diamine oxidase in amniotic tissue was 0. 526 mM in spermine, 0.725 mM in histamine, 0.990 mM in cadaverine, 1. 250 mM in spermidine and 1.316 mM in putrescine.
8. ¢¥Km value of diamine oxidase in chorionic tissue was 0. 676 mM in spermine, 0.980 mM in cadaverine, 1.282 mM in putrescine, 1; 538mM in spermidine and 2.381 mM in histamine.
